Research – In vitro antimicrobial effects of Myristica fragrans essential oil on foodborne pathogens and its influence on beef quality during refrigerated storage

Nutmeg (Myristica fragrans) seed was subjected to the hydro‐distillation method to extract its essential oil (NEO). Its main constituents were α‐pinene (20.16%), sabinene (14.45%), and β‐pinene (13.26%) with great antimicrobial and antioxidant actions. A novel edible coating was then fabricated based on the sage seed mucilage (SSM) and NEO, to ameliorate the quality and shelf‐life of beef slices. The NEO‐loaded SSM coating was able to significantly decrease the population of total viable count, Escherichia coliStaphylococcus aureus, psychrotrophic bacteria, and fungi. Moreover, lipid oxidation of beef slices was remarkably suppressed upon the application of bioactive NEO‐loaded SSM edible coating, as compared with the control by Day 6. The coated beef sample, especially NEO‐rich SSM coated ones perceived a higher consumer acceptance and firmness than the SSM coated and noncoated samples. The SSM edible coating containing NEO could therefore have the potential to control the growth of pathogenic microorganisms and lipid oxidation, and to improve the color stability and sensory properties of meat and meat products.

To stop the spread of disease, it could be used to coat phone screens and keyboards, as well as the inside of catheters and breathing tubes, which are a major source of healthcare-associated infections (HCAIs).

The most well known HCAIs are caused by Clostridioides difficile (C. difficile), methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli (E. coli). They commonly occur during in-patient medical or surgical treatment, or from visiting a healthcare setting and pose a serious health threat, making them a key priority for the NHS to address*.

The research, published today in Nature Communications, is the first to show a light activated antimicrobial coating successfully killing bacteria in low intensity, ambient light (300 Lux), such as that found in wards and waiting rooms. Previously, similar coatings needed intense light (3,000 Lux), like that found in operating theatres, to activate their killing properties.

The new bactericidal coating is made of tiny clusters of chemically modified gold embedded in a polymer with crystal violet — a dye with antibacterial and antifungal properties.

 

Research – Biofilm-inhibition activities of fractions of Senna Siamea (LAM) Irwin & Barneby leaf against Escherichia coli

Escherichia coli has carved its niche in the urinary tract with the formation of a formidable matrix called biofilm. This biofilm is not only recalcitrant to the body’s immune system but also resistant to antibacterial agents. Senna siamea (Lam) Irwin and Barneby is a medicinal plant with established antibacterial effect against planktonic cells of many bacteria. An attempt was made herein to evaluate the effect of its leaf extract and fractions on biofilm of E. coli isolates. Crude extracts of leaf, stem bark and root of this plant were prepared using ethanol as the solvent for the cold extraction. Phytochemical screening was carried out on the three extracts. Two E. coli strains from different antenatal patients attending General Hospital, Kafanchan, Kaduna were donated to us by a researcher from Ahmadu Bello University, Zaria and the reference strain, E. coli, WDCM 00013 (from Germany) were tested for biofilm production using the Congo red method. Antimicrobial susceptibility testing of the crude extracts against the isolates was carried out using the agar diffusion method. The Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC) were determined for the leaf extract of the plant using micro broth dilution and agar diffusion methods respectively. In order to establish the antibiofilm activities of the leaf extract of the plant, sub-inhibitory concentrations (sub-MIC) were used against the test isolates in the remaining assays in the work. Column chromatography backed by thin layer chromatography (TLC) was used to fractionate leaf extract (having the best antibacterial activity) of the plant, using different ratios of a combination of hexane, ethyl acetate and n-butanol as fractionating solvents. MIC and MBC of the leaf extract were and 50 mg/ml respectively. High values of percentage biofilm inhibition were observed against all the bacterial isolates from the antibiofilm assay. Combination of solvents in the increasing order of polarity enhanced the antibiofilm activity of the various fractions of the leaf extract of Senna siamea. In conclusion, further fractionation of Senna siamea leaf extract increases its antibiofilm activities.

Dear Mr. Beach:

The U.S. Food and Drug Administration (FDA) inspected your sprouting operation located at 799 51st Street, Marion, Iowa, from December 31, 2019, to January 9, 2020.  FDA conducted this inspection because sprouts grown at your operation were linked by the state of Iowa Department of Inspections and Appeals, Department of Public Health, and the (b)(4) to an outbreak of human infections with Shiga toxin-producing Escherichia coli O103 (E. coli O103) in the state of Iowa.  This outbreak sickened 22 people between November 21 and December 14, 2019.  Shiga toxin-producing E. coli is a pathogenic bacterium that can cause serious illness in humans, including diarrhea, often with bloody stools.  Although most healthy adults can recover completely within a week, some people can develop a form of kidney failure called hemolytic uremic syndrome (HUS).  HUS is most likely to occur in young children and the elderly.  This condition can lead to serious kidney damage and death.

The U.S. Food and Drug Administration (FDA), along with the Centers for Disease Control and Prevention (CDC), and state and local partners, have collaboratively investigated several outbreaks linked to Jimmy John’s restaurants.  In this letter we detail evidence from five outbreaks, including the most recent outbreak in the state of Iowa during December 2019 of human infections with Escherichia coli O103.  The evidence demonstrates that your corporation, through your franchised Jimmy John’s restaurants, engaged in a pattern of receiving and offering for sale adulterated fresh produce, specifically clover sprouts and cucumbers.

Recommendation

FDA, along with CDC and state and local partners, are investigating an outbreak of 14 illnesses caused by E. coli O103 in Iowa, Illinois, Missouri, Texas and Utah. CDC is reporting that the epidemiologic evidence indicates that clover sprouts from Jimmy John’s restaurants are a likely source of this outbreak.

The CDC is recommending that consumers who have recently eaten clover sprouts on a sandwich from Jimmy John’s restaurants and who are experiencing symptoms of E. coli infection, such as severe stomach cramps, diarrhea, fever, nausea, and/or vomiting, should contact their doctor and report their illness to their local health department. If consumers have a leftover sandwich with clover sprouts from Jimmy John’s, do not eat it and throw it away.

Generally, it is recommended that children, the elderly, pregnant women, and persons with weakened immune systems should avoid eating raw sprouts of any kind.

Jimmy John’s LLC reported that all of its restaurants stopped serving clover sprouts on February 24, 2020. At this time, clover sprouts should not be available at Jimmy John’s.

FDA has activated a team to coordinate this outbreak investigation. This team is currently collecting records and initiating a traceback investigation to determine the source of the clover sprouts. Authorities are investigating whether implicated sprouts have been distributed elsewhere and will continue monitoring for additional illnesses.

Background

On February 21, 2020, the FDA issued a warning letter to Jimmy John’s Franchise LLC for engaging in a pattern of receiving and offering for sale adulterated fresh produce, specifically clover sprouts and cucumbers. On February 24, 2020, Jimmy John’s reported to FDA that they had removed sprouts from their restaurants.

The warning letter lays out evidence from five outbreaks, including recent outbreaks in the state of Iowa during November and December 2019, of human infections with Escherichia coli O103, a Shiga toxin producing Escherichia coli (STEC) linked to sprouts.

These 14 illnesses are new and are not associated with the outbreaks mentioned in the Jimmy Johns warning letter. FDA will continue its investigation and provide updates as more information becomes available.

An E. coli O103 Jimmy John’s clover sprouts outbreak has sickened at least 22 people in Iowa, according to a warning letter the FDA sent to Jimmy John’s and Sprouts Unlimited on February 21, 2020. This outbreak was briefly mentioned by the FDA in a recall notice on December 18, 2019, but there were very few details about the outbreak at that time.

Canada – Silani Mozzarella Ball Recalled in Canada for Generic E. coli

Silani Sweet Cheese Ltd. is recalling Silani Mozzarella Ball for possible generic E. coli contamination. That means this E. coli does not necessarily cause human illness, but it is an indicator of contamination with fecal matter. Generic E. coli is found in the intestines of animals.

The recalled product was sold nationally in Canada at the retail level. This is a Class 2 classification recall, which means “in which the use of, or exposure to, a violative product may cause temporary adverse health consequences or where the probability of serious adverse health consequences is remote.”

The recalled product is Silani Mozzarella Ball, sold in 260 gram packages. The UPC number on the product is 0 65052 51369 4, and the code on the product is Best Before: 2021.01.08.

USDA data on daily shipments of romaine lettuce can be used to determine which production regions are free from contamination during a foodborne illness outbreak. These data, reported by USDA’s Agricultural Marketing Service with a one- or two-day lag, provide essentially real-time information on produce shipped out to retailers. An analysis conducted by the USDA’s Economic Research Service (ERS) demonstrates how this information may allow FDA to rule out an entire production region as the source of contamination.

Leafy greens, including romaine lettuce, are the sixth most commonly consumed type of vegetable in the United States. From May to November, most romaine lettuce in the U.S. comes from California’s Central Coast region; from December to April, most comes from the Yuma, Ariz. region. Among the 29 outbreaks of Shiga-toxin producing Escherichia coli O157:H7 (E. coli) associated with romaine lettuce between 1998 and 2018, illnesses peaked in April and October, which corresponds with the tail end of harvest season in the two main romaine growing regions. In 2017 and 2018, there were three multistate, multinational foodborne illness E. coli outbreaks associated with the consumption of romaine lettuce that occurred at the tail end of either Yuma, Ariz., or California’s Central Coast production seasons. These outbreaks led to a total of 376 illnesses, 158 hospitalizations, and 7 deaths.
